Multimodal imaging platform by CrestOptics and Tomocube

The platform uses 3D holotomographic imaging and fluorescence-based detection, enabling advanced and precise imaging

Fluorescence microscopy developer CrestOptics S.p.A. and holotomography (HT) technology specialist Tomocube have partnered to produce multimodal imaging platform HT-X1 Plus.

The multimodal imaging platform uses high resolution 3D holotomographic imaging alongside fluorescence-based detection to enable advanced and precise label-free biophysical imaging.

“We’ve seen a growing demand in recent years for a solution that combines the label-free imaging of holotomography with the precision of fluorescence-based microscopy,” said Tomocube vice president of customer development Dr Sumin Lee.

“This partnership has enabled us to bring that vision to life with HT-X1 Plus, an industry-first spinning-disk confocal holotomographic instrument that provides unprecedented resolution and sensitivity for 3D biophysical imaging applications.”

This is possible by combining CrestOptics’ spinning disk confocal technology and Tomocube’s HT imaging technology.

HT imaging illuminates a live cell with low power visible light from many angles. It then involves the measurement of the phase delay of the transmitted light. Tomocube’s technology uses the measured refractive index as imaging contrast to allow the visualisation of 3D living cells and tissues without labelling or staining.

The HT-X1 Plus combines Tomocube’s imaging modality with CrestOptics’ to facilitate deeper, more robust insights into molecular distribution within samples without invasive methods.

The precision alignment of HT and fluorescence-based imaging datasets enables researchers to study molecular and phenotypic data together and generate more powerful insights across biophysical imaging applications.

CrestOptics and Tomocube presented HT-X1 Plus at the Spanish and Portuguese Advanced Optical Microscopy Meeting (SPAOM) 2024.
